Colonna: Luna C18 (250 × 3 mm × 5 μm) (Phenomenex, PA, USA);Volume d’iniezione: 2 μL;Mobile phase: acqua/acido formico (99.5:0.5, v/v) (solv. A), acetonitrile/acido formico (99.5:0.5, v/v) (solv. B);Gradiente: 0 min, 0% B, 5 min, 0% B, 15 min, 22% B, 20 min, 100% B, 25 min, 100% B;Flusso: 0.3 mL/min;Rivelatore: range 190-400 nm, cromatogrammi estratti a 245 nm.

Ascorbato totale determinato dopo riduzione di DHA ad ascorbicocon DTT.

Gli estratti vengono addizionati con tampone fosfato 150 mM (pH7.4), lasciati a temp. ambiente per 10 minuti.La reazione colorimetrica avviene aggiungendo i seguentireagenti: acido ortofosforico, FeCl3, TCA, α,α’-dipyridyl in 70%etanolo, e lasciando a 40°C per 40 minuti.

Assorbanza letta a 525 nm.

Determinazione dei fenoli totali

- Metodo Folin-CiocalteuL’estratto in metanolo/acqua di ciascun campione addizionato del reattivo,viene incubato per 8 minuti a temperature ambiente. Si addizionacarbonato di sodio e si lascia a reagire al buio per 2 ore. Si legge quindil’assorbanza a 765 nm.

- Saggio DPPHL’estratto in metanolo/acqua di ciascun campione viene addizionato diDPPH e lasciato al buio per 20 minuti a temperatura ambiente. Si leggequindi l’assorbanza a 518 nm.

Determinazione dell’attività “free radical scavenging” delle molecolebiologicamente attive

- Saggio TEACL’estratto in metanolo/acqua di ciascun campione viene addizionato ditampone fostato 5 mM e addizionato della soluzione radical cationicaABTS•+ . Si legge quindi l’assorbanza a 734 nm.
